NOG マウスの開発
ヒト細胞、ヒト組織をIn vivoで解析できるヒト化マウスは、創薬のための研究材料としての利用のみならず、新薬の前臨床試験やヒト細胞の分化、機能解析等の基礎研究にも有用であり、医療への貢献が充分期待できる実験動物です。
1988年ヒト化マウスのさきがけとして、免疫不全SCIDマウス(Bosma CG et al., Nature 1983)にヒト造血系組織(McCune JM et al., Science 1988)、もしくはヒト末梢リンパ球(Mosier DE, et al., Nature 1988)を移植したいわゆるSCID-huマウスが開発されました。その後、SCIDマウスを改良したNOD/scidマウスの開発によって、SCID マウスを上回るヒト細胞の生着が可能となりましたが、in vivo解析モデルとしては不十分な点が多く残されておりました。
当研究所では、NOD/scidマウスと数種のサイトカインレセプター共通ドメインである IL-2レセプターγ鎖ノックアウト (IL2RγKO) マウス(Ohbo K et al., Blood 1996)を組み合わせることにより、極めて重度な複合型免疫不全を呈する“NOG (NOD/Shi-scid, IL-2Rγnull) マウス”(Ito M et al., Blood 2002)の樹立に成功しました。
NOGマウスはヒト細胞、ヒト組織の生着性がNOD-scidマウスと比べて著しく高く、ヒト癌細胞やヒト肝臓細胞を高率に生着させることが可能であります。またヒト造血幹細胞移植後に、NOD-scidマウスでは見られなかったヒトT細胞の分化が末梢リンパ組織で認められることから、ヒト免疫系モデルマウスとしての需要も高まってきております。
